Design: Our collection of libraries built for sequencing on the Illumina HiSeq2000 consisted of short-insert (200 bp), and mate-pair libraries (insert sizes: 3, 8, and 10 kbp) all sequenced using 100 bp paired-end reads. The resulting coverage was approximately 90x: 40x of overlapping pairs, 40x of 3 kb, and 10x 8–10 kb.

We have developed a sequencing and assembly strategy use short-read Illumina genome sequencing to perform a de-novo assembly of the P. kingsleyae genome at 90x depth. A fully sequenced Paramormyrops genome will represent the first Osteoglossomorph genome, which is of central interest to those studying the evolution of vertebrate genomes. Osteoglossomorphs are the most ancestral group of teleost fishes, and are the first group to branch from other teleosts following the hypothesized teleost-specific whole genome duplication event. In addition to its utility for genome biologists, this would represent the first mormyrid electric fish genome, and second electric fish genome to be sequenced. This will provide the broader electric fish community with a powerful genomic resource for studies in endocrinology, neurobiology, and evolutionary biology. For the purposes of this specific project, the Paramormyrops reference genome will enhance the quality of comparative transcriptome analysis, and enable the comparison of low-depth genome resequencing sequences acquired from P. kingsleyae EOD types.
